4.  Cell culture system for screening of bioactive compounds from Streptomycetes and      environmental fungi

Objectives: To evaluate cytotoxicity and stimulatory effect of bioactive compounds.

          Nowadays, public health problem are increased due to an increase of drug resistant pathogens. Misuse of existing antibiotics in human results in the development of microbs to resist antimicrobial drugs. Our laboratory corroborates with Professor Watanalai Panbangred, Dr. Arjaree Nilawongse and Dr. Thipa Asvaruk (Department of Biotechnology) to screen new bioactive compounds from Streptomycetes and fungi.¬†Two types of cells including human lung carcinoma cell line (A549) and mouse macrophage cell line (RAW264.7) have been used to evaluate cytotoxicity and stimulatory effect of bioactive compounds which have antimicrobial activity. Cytotoxicity is analyzed by lactate dehydrogenase assay and MTT assay. The concentration-effect relationships and IC50 are determined. Cellular morphological changes are observed under an inverted microscope and cell viability is determined using trypan blue dye-exclusion. We hope that new bioactive compounds can solve drug resistance problem. Furthermore, other compounds may be found such as anti-cancer product and natural compounds as immunomodulators.
